Mesenchymal stem cell-derived extracellular vesicles as probable triggers of radiation-induced heart disease

چکیده

Abstract Background Radiation-induced heart disease has been reported, but the underlying mechanisms remain unclear. Mesenchymal stem cells (MSCs), also residing in heart, are highly susceptible to radiation. We examined hypothesis that altered secretion of extracellular vesicles (EVs) from MSCs is trigger radiation-induced disease. Methods By exposing human placental tissue-derived 5 Gy γ-rays, we then isolated EVs culture medium 48 h later and evaluated changes quantity quality after radiation exposure. The biological effects irradiated on HUVECs H9c2 were examined. Results Although amount size distribution did not differ between nonirradiated MSCs, miRNA sequences indicated many upregulated or downregulated miRNAs EVs. In vitro experiments using HUVEC showed MSC-EVs decreased cell proliferation ( P < 0.01), increased apoptosis DNA damage. Moreover, impaired tube formation induced calcium overload cells. Conclusions released show profiles harmful cells, which provides new insight into mechanism radiation-related risks.

Mesenchymal Stem Cell‐Derived Extracellular Vesicles as Mediators of Anti‐Inflammatory Effects: Endorsement of Macrophage Polarization

Mesenchymal Stem Cells (MSCs) are effective therapeutic agents enhancing the repair of injured tissues mostly through their paracrine activity. Increasing evidences show that besides the secretion of soluble molecules, the release of extracellular vesicles (EVs) represents an alternative mechanism adopted by MSCs. Since macrophages are essential contributors toward the resolution of inflammatio...

Mesenchymal stem cell-derived extracellular vesicles ameliorate inflammation-induced preterm brain injury

OBJECTIVE Preterm brain injury is a major cause of disability in later life, and may result in motor, cognitive and behavioural impairment for which no treatment is currently available. The aetiology is considered as multifactorial, and one underlying key player is inflammation leading to white and grey matter injury. Extracellular vesicles secreted by mesenchymal stem/stromal cells (MSC-EVs) h...

Mesenchymal Stem/Stromal Cell-Derived Extracellular Vesicles and Their Potential as Novel Immunomodulatory Therapeutic Agents

Extracellular vesicles (EVs), such as exosomes and microvesicles, have been identified as mediators of a newly-discovered intercellular communication system. They are essential signaling mediators in various physiological and pathophysiological processes. Depending on their origin, they fulfill different functions. EVs of mesenchymal stem/stromal cells (MSCs) have been found to promote comparab...

Pathogenic or Therapeutic Extracellular Vesicles in Rheumatic Diseases: Role of Mesenchymal Stem Cell-Derived Vesicles

Extracellular vesicles (EVs) are important mediators of cell-to-cell communication pathways via the transport of proteins, mRNA, miRNA and lipids. There are three main types of EVs, exosomes, microparticles and apoptotic bodies, which are classified according to their size and biogenesis. EVs are secreted by all cell types and their function reproduces that of the parental cell. They are involv...
