Mesenchymal stem cell-derived extracellular vesicles as probable triggers of radiation-induced heart disease

Abstract Background Radiation-induced heart disease has been reported, but the underlying mechanisms remain unclear. Mesenchymal stem cells (MSCs), also residing in heart, are highly susceptible to radiation. We examined hypothesis that altered secretion of extracellular vesicles (EVs) from MSCs is trigger radiation-induced disease. Methods By exposing human placental tissue-derived 5 Gy γ-rays, we then isolated EVs culture medium 48 h later and evaluated changes quantity quality after radiation exposure. The biological effects irradiated on HUVECs H9c2 were examined. Results Although amount size distribution did not differ between nonirradiated MSCs, miRNA sequences indicated many upregulated or downregulated miRNAs EVs. In vitro experiments using HUVEC showed MSC-EVs decreased cell proliferation ( P < 0.01), increased apoptosis DNA damage. Moreover, impaired tube formation induced calcium overload cells. Conclusions released show profiles harmful cells, which provides new insight into mechanism radiation-related risks.
